A Study on the Analysis fo Aflatoxin B1 in Human Sera by ELISA

Abstract
|
|
|
We measured the contents of Aflatoxin B in the serum of 111 persons which include liver disease and normal control during the period of Nov.1 1989 to Feb.30 1990 at the Gyeong-Sang national University Hospital. There were 47 cases of liver
cirrhosis, 18
cases of hepatoma and 46 cases of normal control. The analysis was performed by competitive direct ELISA referred to Pestka et al.
1) Rabbit antiserum against Aflatoxin B oxime-BSA reached the peak level of antibody titer on 13 weeks after immunization of antigen and showed high titer at 1,000 times dilution in rabbit A.
2) Antiserum produced by Aflatoxin B showed strong cross reaction to Aflatoxin B(11.8%) compared to Aflatoxin B and lesser extent with Aflatoxin G1(28%) and G2(36%) and almost none to Aflatoxin M(1%).
3) The standard curve of Aflatoxin B1 obtained by addition of methanol-H2O(70 ; 30) after spiking of standard Aflatoxin B1 into sterile blood for one night showed slight higher value than standard curve of standard Aflatoxin B1 treated with
methanol
but it didn't affect to the results.
4) Among 111 cases, 2 cases of hepatoma(11.1%) and 1 cases of liver cirrhosis(12.12%) showed positive level and the level of detected titer were H-17 : 1.83±0.20 ng/ml, the highest level, H-4 : 0.68±0.01 ng/ml, L-22 : 0.52±0.12 ng/ml,
respectively.
5) The contents of Aflatoxin B1 were not influenced by specific districs or diseases.
